An in vitro investigation of the effectiveness of commercially available alpha-anylase inhibitors

The purpose of this study was to identify factors which might render alpha-amylase inhibitors ineffective. An in vitro colorimetric investigation was performed in which porcine pancreatic alpha-amylase, preincubated with inhibitor, was added to a starch solution. The preincubation time, pH, and temperature of the starch solution, along with the type of starch used, were manipulated. It was found that when preincubation time was decreased, the inhibitor's effectiveness decreased, and when the pH was increased to 8.2 the inhibitor was virtually ineffective.
