A comparison of the effects of diabetes mellitus and ethane-1-hydroxy-1, 1-diphosphonic acid on plasma and bone calcium metabolism in the rat

Plasma and bone calcium levels were measured in both alloxan diabetic and Ethane-1-hydroxy-1, 1-diphosphonic acid (EHDP) treated rats. The investigation was based on the hypothesis that both diabetes and treatment with EHDP inhibits the l-α hydroxylase enzyme necessary for the conversion of 25-OH D2 to 1,25 (OH)2 D3. This may alter calcium metabolism and normal bone mineralization in rats. Plasma calcium levels were unaltered in all groups (control, uncontrolled diabetic, controlled diabetic, and EHDP treated) indicating that the non-overnight fasted rat can maintain calcium homeostasis regardless of the treatment status. Uncontrolled diabetes and treatment with EHDP showed parallel results with a decrease in bone mass and an increase in bone calcium concentration. These findings show that diabetes and EHDP may be inhibiting 1,25 (OH)2 D3 action on resorption of bone, and the bones are losing mineral mass other than calcium. Whether the lost mineral is phosphorus or some other mineral is not known. Our study indicates a similarity in physiochemical action for uncontrolled diabetes and treatment with EHDP on altering bone metabolism in the rat.
